Automated Determination of Fluvoxamine in Plasma by Column-Switching High-Performance Liquid Chromatography

Abstract A column-switching system with high-performance liquid-chromatographic separation and ultraviolet detection is described for automated determination of fluvoxamine in human plasma or serum. Samples were injected and the drug was retained in a clean-up column [20 x 4.6 mm (i.d.)] filled with C8 reversed-phase material (10-micron particles). H-Point Curve Isolation Method for Coupled Liquid Chromatography and UV−Visible Spectrophotometry

The H-point curve isolation method (HPCIM) for the detection of unknown interferences in chromatography is proposed. The method allows one to estimate the UV-vis spectra of interfering species in a sample as well as to test the purity of the chromatographic peaks. Besides the detection of the unknown interferences in a sample, this method allows one to calculate the concentration of an analyte in the presence of unknown compounds. To illustrate the reliability of the proposed method, samples of diuretics and amphetamines have been analyzed by normal- and reversed-phase high-performance chromatography.

Gas-liquid chromatographic analyses

Abstract The gas chromatography of mixtures of n -alkyl acetates (CH 3 COOR, R = C 1 —C 8 ) and methyl esters of aliphatic n -carboxylic acids (R′COOCH 3 , R′ = C 1 —C 8 ) and certain of their monochlorinated derivatives has been studied on Carbowax 20M and SE-30 glass capillary columns under the same operating conditions. The separation of the isomeric monochlorinated esters was complete on Carbowax 20M, whereas on SE-30 the peaks of 6- and 7-chlorooctyl acetates and methyl (ω − 1)- and (ω − 2)-chlorooctanoates and -nonanoates partly overlapped. The complete separation of the mixtures could not be achieved, however, on Carbowax 20M, in spite of the use of various operating conditions. The …

Highly efficient separation of amines by electrokinetic chromatography using resorcarene-octacarboxylic acids as pseudo-stationary phases

Abstract Resorcarene-octacarboxylic acids, macrocyclic molecules built up by four alkylidene-bridged resorcinol units, were synthesized and used as pseudostationary phases in electrokinetic chromatography (EKC). Resorcarenes provide a stable structure and good solubility in electrolytes even with organic modifiers. The high electrophoretic mobility of the resorcarene-octacarboxylic acids introduced here as pseudostationary phases is based on the eight partly deprotonated carboxylic groups. The interactive modes of High Performance Liquid Chromatography (HPLC) of proteins provide a platform for the construction of a multidimensional HPLC system coupled to mass spectrometry. We present a system composed of both anion and cation exchanger columns, in the first dimension, and n-octadecyl bonded 1.5 μm nonporous silica columns in the second dimension. Both columns are operated under gradient conditions. A system suitability test with standard proteins showed that the total analysis can be performed within about 20 minutes. Liquid chromatography with single columns often does not succeed in the analysis of complex samples, in terms of resolution and analysis time. A relatively simple solution to enhance chromatographic resolution is the modulation of the stationary phase through the serial coupling of columns. Glass capillary gas chromatography of chlorinated methyl acetates, propanoates and butanoates on Carbowax 20M and SE-30 columns

The gas chromatography of all chlorinated methyl acetates, methyl propanoates and methyl mono- and dichlorobutanoates has been studied on Carbowax 20M and SE-30 glass capillary columns under various running conditions. The order of elution on a non-polar column was largely determined by the boiling point of esters, whereas on a polar column it was much influenced by the structure of compounds. Complete separation of the combined mixture of all 27 compounds could not be achieved, however, methyl 3,3-dichlorobutanoate was the only ester overlapped on both columns in spite of the various column temperatures used. Gel permeation Chromatography in coiled columns

Whereas in gas chromatography the use of coiled columns is the usual procedure, in liquid chromatography the use of straight columns is favoured. Bent columns give an additional contribution to peak spreading which is of greater importance in liquid chromatography than in gas chromatography owing to smaller diffusion coefficients. The low linear velocities of the mobile phase used in gel permeation chromatography enable conditions to be found where the additional contribution of bending to peak spreading is small.
